cellstructuremech:radial_microtubule_organizing_center · Cytoskeleton · Multiprotein complex · Proposed

A transient, Hrs1/Mcp6-dependent, gamma-tubulin-enriched amorphous microtubule-organizing assembly at the cytoplasmic side of the spindle pole body during Schizosaccharomyces pombe meiotic prophase, distinct from the spindle pole body core, that organizes the radial microtubule array used in horsetail nuclear movement. [DOI:10.1016/j.cub.2012.02.042]

Gamma-tubulin localization at the S. pombe radial organizing center. A shows immunogold-labeled TEM serial sections of synchronized meiotic cells with wild-type hrs1; B-C model those labels and the ribosome-excluding amorphous region. D shows hrs1-deletion sections and E their model, with more dispersed gamma-tubulin labels and nonradial microtubules. In the models, yellow marks gamma-tubulin labels, green microtubules and blue the SPB; C highlights the amorphous region. Bars are 100 nm. Model surfaces are interpretations, not additional micrographs.

Hrs1/Mcp6-like meiotic organizing proteinProteinpart ofReviewed label only (reviewed name; no accession asserted)mcp6, hrs1, mug3ESSENTIALLocalized constituent required for the recognizable meiotic rMTOC in S. pombe. A structural-scaffold role is supported as a model; a purified molecular bridge and its stoichiometry are not resolved.
  • DOI:10.1016/j.cub.2012.02.042 Immunogold labeling places Hrs1 in the faintly electron-dense, ribosome-excluding region outside the SPB core. hrs1 deletion removes this recognizable region and normal radial geometry while retaining some nonradial SPB-associated microtubules. Essentiality concerns rMTOC formation, not cell viability or all microtubule polymerization.
Gamma tubulinProteinpart ofInterPro:IPR002454gtb1, tug1UNKNOWNGamma-tubulin class enriched in the radial organizing region; local enrichment does not specify a complete ring complex or demonstrate sufficiency for rMTOC assembly.
  • DOI:10.1016/j.cub.2012.02.042 Results section 2.3 and Figure 3 establish gamma-tubulin enrichment in the rMTOC. This study does not separately test whether gamma-tubulin removal abolishes the amorphous scaffold, so assembly essentiality is left UNKNOWN.

  • DOI:10.1016/j.cub.2012.02.042 Figure 3 detects S. pombe gamma tubulin in the rMTOC with GTU88 immunogold labeling; the signal becomes more dispersed around the SPB in hrs1 deletion. Hrs1-dependent radial center assembly (ASSEMBLY)

S. pombe meiotic prophase. Hrs1 loss tests assembly dependence and local gamma-tubulin enrichment; constituent localization is distinct from a purified binding or nucleation reaction. The HAA is the output array, not an rMTOC constituent. Nuclear movement additionally requires motor machinery.
