- Source: DSMZ | Link: https://www.dsmz.de/microorganisms/medium/pdf/DSMZ_Medium119b.pdf Superseded historical provenance (not a current formulation claim): Commercial Product: LB Medium (Luria-Bertani, Miller formulation) Supplier: Multiple suppliers (Difco, Sigma, etc.) Catalog: Multiple (L3522 Sigma, 244520 BD) Source: https://www.laboratorynotes.com/preparation-of-luria-bertani-lb-miller-broth/ Accessed: 2026-03-28 Constituent composition researched from product specification. Archive all prior ingredient nodes, including false LB expansion, combined yeast amounts and copied or chemically inexact annotations, in append-only history. New nodes follow this exact source's stage and label; other unchanged chemical annotations are preserved, not recertified. Withdraw the unsupported K119/DSMZ119b exact-duplicate edge: K119 links original DSMZ119, whereas DSMZ119b includes methanol and selenite-tungstate and different bicarbonate, water and preparation. Their old copied ingredient signatures are not source evidence of identity. Source-scoped recipe inputs supersede unsupported flattened concentrations and false LB identities. All prior history and permanent IDs remain unchanged; unquantified quantities remain unknown. No taxa-media association is added.

Preparation
- Dissolve ingredients except methanol, bicarbonate, cysteine and sulfide. Adjust pH of medium to 7.2 and sparge with 100% N2 gas for 30 - 45 min to make it anoxic. Then dispense medium under same gas atmosphere into anoxic Hungate-type tubes to 30% v/v of their volume and autoclave. Add methanol (from 15% v/v stock solution), cysteine and sulfide from sterile anoxic stock solutions prepared under 100% N2 gas and bicarbonate from a sterile anoxic stock solution prepared under 80% N2 and 20% CO2 gas mixture. Prior to use check pH of complete medium and adjust to 7.6, if necessary.
- After inoculation, add sterile 80% H2 and 20% CO2 gas mixture to 0.5 bar overpressure. After growth becomes visible overpressure of 80% H2 and 20% CO2 gas mixture can be increased to 1 bar.
- Note: Use 10% (v/v) as inoculum.
