- Source: DSMZ | Link: https://www.dsmz.de/microorganisms/medium/pdf/DSMZ_Medium265.pdf Original-source preparation scope: mineral salt base plus separately prepared yeast-extract, sucrose, sulfur, polar-lipid and sulfide additions under 100% nitrogen. Historical aerobic/import annotations are not endorsed. Basal rates remain provider-declared 1000 ml normalizations only. The original PDF supplies stock strengths and batch doses; its lipid range differs from the API's modified protocol. Neither source establishes a measured final receiving volume. Chemical mappings, parent/variant links and prior curation history are preserved, not independently revalidated.

Source batch input: 10 g powdered sulfur, sterilized separately by steaming for 3 h on each of three successive days and added to the autoclaved mineral salt preparation. The former 10 g/L is not retained as a final-medium concentration; final volume is unspecified. |
Preparation
- Adjust final pH to 5.2.
- Prepare the medium anaerobically under 100% nitrogen. The following constituents are prepared separately and added to the autoclaved mineral salt solution: Yeast extract (20 ml of 10% w/v solution)-boiled for few minutes, not autoclaved; sucrose (20 ml of 10% w/v solution)-filter-sterilized; sulfur (10 g)-sterilized by steaming for 3 h on each of three successive days; polar lipid fraction (6 - 12ml)-prepared as described by W. Zillig et al. (1983), Syst. Appl. Microbiol. 4: 79 - 87; Na2S x 9 H2O (10 ml of 3% w/v solution)-autoclaved under nitrogen atmosphere.
- Sparge medium with 100% N2 gas mixture to make anoxic.
- Autoclave the mineral salt preparation at 121 °C for 15 min; prepare the supplements separately as specified.
- after autoclaving add the following sterile solutions:
- Note: lipid fraction can be prepared from fresh or freeze-dried biomass. Cells are disrupted by autoclaving or sonication, then filter-sterilized. Use 5 g wet or 0,2 g dried biomass in 10 ml water or buffer, and use 10ml/l of the sterile-filtered extract. Prepare under N2 conditions. Modified from W. Zillig et al. (1983), Syst. Appl. Microbiol. 4: 79 - 87

| Distilled water | 1 L | 1000 ml distilled-water input per mineral-base batch, not a final-volume instruction. |
1000 ml distilled water is a batch input, not final-volume makeup. The retained basal g/L values are provider-reported rates on the API's 1000 ml basis, not measured final stock or complete-medium concentrations. No base addition volume is inferred.
Yeast extract stock (10% w/v)
Dose: 0.02 L
| Ingredient | Concentration | Notes |
|---|---|---|
| Yeast extract | 10 PERCENT_W_V | 10% w/v in the separately prepared stock, not the former imported final-medium rate. Original source: 20 ml stock contains 2 g yeast extract; the API's 20 mg/l and former 0.02 g/L disagree. The PDF calls for boiling, not autoclaving; its instruction does not add a separate yeast filtration step. |
| Water | Stock solvent according to the API attribute; no fixed carrier volume is specified. |
Original-recipe batch addition, not liters per liter of final medium. Stock strength and batch dose are separate claims; the stock solvent amount and final receiving volume are not inferred.
Sucrose stock (10% w/v)
Dose: 0.02 L
| Ingredient | Concentration | Notes |
|---|---|---|
| Sucrose | 10 PERCENT_W_V | 10% w/v in the separately prepared stock, not the former imported final-medium rate. Receiving-medium concentration is unresolved because its final volume is not established. |
| Water | Stock solvent according to the API attribute; no fixed carrier volume is specified. |
Original-recipe batch addition, not liters per liter of final medium. Stock strength and batch dose are separate claims; the stock solvent amount and final receiving volume are not inferred.
Na2S x 9 H2O stock (3% w/v)
Dose: 0.01 L
| Ingredient | Concentration | Notes |
|---|---|---|
| Na2S x 9 H2O | 3 PERCENT_W_V | 3% w/v in the separately prepared stock, not the former imported final-medium rate. |
Polar lipid aqueous suspension
Dose: 0.006-0.012 L
| Ingredient | Concentration | Notes |
|---|---|---|
| Polar lipid fraction | Source material: aqueous polar-lipid suspension prepared from Thermoproteus tenax (DSM 2078) or another archaebacterium. Composition, extraction yield and lipid mass concentration are not specified. Former mass/percentage encoding is withdrawn. |
Original PDF batch dose: 6-12 ml. The API alternatively lists 10 ml/l and a modified biomass extraction procedure; that version is retained in evidence, not silently substituted for the original range or used to calculate final lipid concentration.
