Notes
Source: DSMZ | Link: https://www.dsmz.de/microorganisms/medium/pdf/DSMZ_Medium857.pdf Superseded historical provenance (not a current formulation claim): Commercial Product: LB Medium (Luria-Bertani, Miller formulation) Supplier: Multiple suppliers (Difco, Sigma, etc.) Catalog: Multiple (L3522 Sigma, 244520 BD) Source: https://www.laboratorynotes.com/preparation-of-luria-bertani-lb-miller-broth/ Accessed: 2026-03-28 Constituent composition researched from product specification. False LB constituent expansion is withdrawn. Source-specific amounts and preparation boundaries supersede flattened concentrations. Existing chemical annotations and earlier history are preserved, not recertified; no organism-growth association is added. Original DSMZ 857 protocol explicitly prepares one liter final medium with 900 ml water and 2 ml glycerol, then adds 100 ml ADC after cooling. Do not autoclave the added enrichment or replace its 0.3 mg catalase with another recipe's OADC amount.

13 Distilled water 0.9 L Source preparation input, 0.9 L; water is 900 ml and glycerol is 2 ml. The API table's 1000 ml water row conflicts with its preparation prose and the original PDF; this input follows the explicit preparation, not a sum of component volumes.
14 Glycerol 0.002 L Source preparation input, 0.002 L; water is 900 ml and glycerol is 2 ml. The API table's 1000 ml water row conflicts with its preparation prose and the original PDF; this input follows the explicit preparation, not a sum of component volumes.

Preparation

  1. Solution g/liter (final solution) 4.7 g/l. Final pH at 25°C 6.6 ± 0.2.
  2. To prepare one liter of final medium, suspend the amount of dehydrated powder indicated under FORMULAE-Solution (and on the package label) in 900 ml distilled and deionized water containing 2 ml glycerol and heat to boiling to dissolve completely. Sterilize in the autoclave for 10 or 15 min at 15 Ibs pressure (121°C), per label copy. Cool to 50-55°C.
  3. Aseptically add 100 ml of the appropriate ADC Enrichment and mix throughly.
