Preparation

  1. Mix 44.0 g Columbia blood agar base (BD-Difco) with 950.0 ml distilled water.
  2. Autoclave and cool to about 70 C.
  3. Aseptically add 50.0 ml sterile defibrinated sheep blood and keep at 70 C for 15 min.
  4. Cool to about 50 C.
  5. Mix and quickly dispense into sterile petri dishes.
