The exact recipe specifies albumin, not LB broth. Remove only the three reviewed LB supplier nodes and restore albumin at its source-defined preparation stage. Genuine yeast extract and unrelated annotations are preserved.

Preparation

  1. Mix components except yeast extract, bovine serum albumin and Na2S·9H2O.
  2. Bring to a boil for 5-10 sec and flush with N2.
  3. Adjust pH to 7.6 at 80-90C.
  4. After cooling to room temperature, dispense the medium into serum bottles under a N2 atmosphere, seal with butyl rubber stoppers and autoclave.
  5. Separately autoclave yeast extract as a 10% solution and neutralized Na2S·9H2O as a 5% solution under a N2 atmosphere.
  6. Filter-sterilize bovine serum albumin as a 10% (w/v) solution under a N2 atmosphere.
  7. Before inoculation, aseptically and anaerobically add the yeast extract, bovine serum albumin and neutralized Na2S·9H2O solutions.

Solutions

10% yeast extract solution

Stock composition
IngredientConcentrationNotes
Yeast extract Source batch input: 1 g. Final preparation volume is not independently established; the unsupported g/L assertion is withdrawn, not replaced by an assumed one-liter concentration. This is the contribution specified for the receiving medium batch, not the mass for preparing an unspecified volume of stock. Source qualifier: BD-Difco.
Separate stock preparation added aseptically and anaerobically under N2 before inoculation. The source table mass is its contribution to the receiving batch, not a mass for preparing an unspecified stock volume. The stock percentage basis and fixed dose are unspecified.

10% (w/v) bovine serum albumin solution

Dose: 0.01 L

Stock composition
IngredientConcentrationNotes
Bovine serum albumin 10 PERCENT_W_V Source batch input: 1 g. Final preparation volume is not independently established; the source batch amount is retained without asserting a final concentration. This is the contribution specified for the receiving medium batch, not the mass for preparing an unspecified volume of stock. Explicit albumin-stock strength is 10% (w/v) = 10 g per 100 ml = 100 g/L. This is not the receiving-medium concentration.
Separate stock preparation added aseptically and anaerobically under N2 before inoculation. The source table mass is its contribution to the receiving batch, not a mass for preparing an unspecified stock volume. Derived receiving-batch dose: source contribution 1 g / explicit stock strength (10 g / 0.1 L = 100 g/L) = 0.01 L (10 ml). Both inputs describe this recipe's separately filtered albumin stock. No final-medium volume is needed or inferred; the dose is calculated, not directly printed in the source.

5% neutralized sodium sulfide nonahydrate solution

Stock composition
IngredientConcentrationNotes
Na2S x 9 H2O Source batch input: 0.5 g. Final preparation volume is not independently established; the unsupported g/L assertion is withdrawn, not replaced by an assumed one-liter concentration. This is the contribution specified for the receiving medium batch, not the mass for preparing an unspecified volume of stock.
Separate stock preparation added aseptically and anaerobically under N2 before inoculation. The source table mass is its contribution to the receiving batch, not a mass for preparing an unspecified stock volume. The stock percentage basis and fixed dose are unspecified.
