- Notes
- Source: https://togomedium.org/medium/M2410 Superseded historical provenance (not a current formulation claim): Commercial Product: LB Medium (Luria-Bertani, Miller formulation) Supplier: Multiple suppliers (Difco, Sigma, etc.) Catalog: Multiple (L3522 Sigma, 244520 BD) Source: https://www.laboratorynotes.com/preparation-of-luria-bertani-lb-miller-broth/ Accessed: 2026-03-28 Constituent composition researched from product specification. The false LB constituent expansion is withdrawn in favor of this source's bovine serum albumin input. Existing unrelated chemical annotations are preserved, not recertified. This repair does not assert equivalence with other BSK formulations or add organism-growth evidence. 9. Incubate at 34-37°C *Hamilton, Montana, is at an altitude of 1,200m. At lower elevations the containiers may need to be filled to a higher level.

Preparation
- 1. After detergent cleaning, all glassware is rinsed thoroughly with glass-distilled water and then autoclaved.
- 2. To 900ml of glass-distilled water are added 100ml of 10X concentrate of CMRL 1066 without glutamine (Gibco Laboratories, GrandIsland, NY, No.330-1540).
- Add the source-listed ingredients to the diluted CMRL 1066. The provider requests an addition order, but its item array is not accepted as that order; exact addition order remains unresolved here.
- 4. Adjust pH of medium at 20-25°C to 7.6 with 1 N NaOH.
- 5. Add 200ml of 7 percent gelatin (Difco) which had been dissolved in boilinig water.
- 6. Sterilize by filtration with air pressure (0.2 μm nitrocellulose, Millipore Corp., Bedford, MA). Store medium at 4°C.
- 7. Before use, add unheated rabbit serum(\"trace hemolyzed\"; Pel-Freez Biologicals, Inc., Rogers, AR) to a final concentration of 6 percent.
- 8. Dispense to glass or polystyrene tubes or bottles. Fill containers to 50 to 90 percent capacity and captightly.*
Solutions
10X concentrate of CMRL 1066 without glutamine (Gibco Laboratories, GrandIsland, NY, No.330-1540)
Dose: 0.1 L
Properties: Defined component, Complex component
Source dose: 100 ml = 0.1 L of the specified 10X CMRL 1066 without glutamine. No product composition or final-medium g/L is inferred.
NaOH solution (1 N)
| Ingredient | Concentration | Notes |
|---|---|---|
| NaOH | Properties: Defined component, Inorganic compound, Solution Source stock strength is 1 N, not an asserted molarity or final-medium concentration. Titrate to pH 7.6 at 20-25 C before gelatin/serum additions; no fixed dose is given. |
Gelatin solution (7 percent; basis unspecified)
Dose: 0.2 L
| Ingredient | Concentration | Notes |
|---|---|---|
| Gelatin | Difco gelatin stock: 7 percent, with no stated w/v, w/w or v/v basis. No numeric stock mass concentration is asserted. |
Unheated rabbit serum
Before use, add unheated rabbit serum (trace hemolyzed; Pel-Freez Biologicals) to a final 6 percent. Percentage basis and fixed batch dose are not stated; neither is guessed.
Target organisms
No target organisms are recorded.
