Preparation
- Add components to distilled water and bring volume to 1.0 L. Adjust pH 8.0 with NaOH. Autoclave under a N2 atmosphere. After cooling, add anaerobically the following solutions (autoclaved or *filter--sterilized):
- Readjust pH to 8.0, if necessary.
Solutions
Basal preparation at the 1 L makeup stage
| Ingredient | Concentration | Notes |
|---|---|---|
| Distilled water | Role: Solvating media; Properties: Defined component, Simple component, Inorganic compound [Merged 2 duplicates: 1.0, 25.0] Water is the makeup reagent for the explicitly stated 1.0 L basal stage, not a fixed 1 L water input or a final supplemented-medium volume. Its actual addition amount is unspecified. The historical duplicate-summing note combined basal makeup and reducing-stock water; that interpretation is superseded. | |
| MgSO4・7H2O | 8.8 G_PER_L | Role: Mineral source; Properties: Defined component, Inorganic compound, Simple component Source basal batch amount: 8.8 g; explicitly made up to 1.0 L at this stage. Calculation: 8.8 g / 1.0 L = 8.8 g/L. This concentration applies at basal makeup, before subsequent pH adjustment, heating and supplements; it is not a final supplemented-medium rate. |
| Yeast extract | 10 G_PER_L | Role: Nutrient source; Properties: Undefined component Source basal batch amount: 10 g; explicitly made up to 1.0 L at this stage. Calculation: 10 g / 1.0 L = 10 g/L. This concentration applies at basal makeup, before subsequent pH adjustment, heating and supplements; it is not a final supplemented-medium rate. |
| NaCl | 75 G_PER_L | Role: Mineral source; Properties: Defined component, Simple component, Inorganic compound Source basal batch amount: 75 g; explicitly made up to 1.0 L at this stage. Calculation: 75 g / 1.0 L = 75 g/L. This concentration applies at basal makeup, before subsequent pH adjustment, heating and supplements; it is not a final supplemented-medium rate. |
| KCl | 1 G_PER_L | Role: Mineral source; Properties: Defined component, Inorganic compound, Simple component Source basal batch amount: 1 g; explicitly made up to 1.0 L at this stage. Calculation: 1 g / 1.0 L = 1 g/L. This concentration applies at basal makeup, before subsequent pH adjustment, heating and supplements; it is not a final supplemented-medium rate. |
| Trypticase peptone (BD-BBL) | 10 G_PER_L | Properties: Undefined component Source basal batch amount: 10 g; explicitly made up to 1.0 L at this stage. Calculation: 10 g / 1.0 L = 10 g/L. This concentration applies at basal makeup, before subsequent pH adjustment, heating and supplements; it is not a final supplemented-medium rate. |
| NaOH | Properties: Defined component, Inorganic compound, Solution Source pH-adjustment reagent with no fixed dose. Removed the unsupported VARIABLE placeholder. | |
| N2 | Properties: Gas Source atmosphere context, not a fixed gas input or dissolved concentration. Removed the unsupported VARIABLE placeholder. |
This is the explicitly made-up basal preparation before later supplements. Composition rates apply at the 1.0 L makeup stage only. No fixed dose of the subsequently titrated/heated basal preparation or final supplemented-medium volume is asserted.
