# Ingredient Concentration Ontology Notes
1 Modified Wolfe's solution (see Medium No. 628 ) 1000 ML_PER_L
2 Trace minerals (see Medium No. 151 ) 10.0 ML_PER_L

Preparation

  1. Mix components and autoclave.
  2. Aseptically add per liter the following solutions from sterile stocks (autoclaved or *filter-sterilized): Check final pH to be 6.0-6.5.
  3. Dispense into culture vessels, replace the gas phase with a N2-CO2-O2 (79:20:1, v/v/v) gas mixture, and seal with butyl rubber stoppers.
  4. Modified Wolfe's Mineral solution: Trace minerals: Dissolve nitrilotriacetic acid and adjust pH to 6.5 with KOH.
  5. Then proceed to add minerals.
  6. Adjust final pH to 7.0.
