protocatechuate degradation I (meta-cleavage pathway)
MetaCyc:P184-PWY
The complete MetaCyc 27.0 eight-member protocatechuate 4,5-cleavage definition proceeds through a semialdehyde hemiacetal, pyrone dicarboxylate, oxalomesaconate, the source-defined E,E enol and stereochemically unspecified CHA to oxaloacetate and pyruvate, followed by oxaloacetate decarboxylation. This is a source-defined composite with enzyme and taxon assertions scoped individually; it is not an experimentally reconstituted eight-step route in one strain. Modern LigU/LigJ reactions use distinct KCH and stereospecific S-CHA and are not substituted under the historical native reaction IDs. Taxa also include the explicitly scoped enzyme-source contexts; KT2440 gallate-module activities do not establish complete pathway occurrence in that strain.

4-carboxy-4-hydroxy-2-oxoadipate aldolaseMetaCyc:CPLX-2865experimental construct: Purified recombinant SYK6 LigK from pETK in E. coli BL21(DE3), observed as a homohexamer. The measured N-terminus lacks the initial native methionine; unreported vector-derived residues are not inferred absent. Qualification evidence
| has_cofactor The source annotates Mg as a LigK cofactor. In the reported recombinant assays, CHA activity with 1 mM Co, Zn, Ca or Mn was 85%, 65%, 20% or 0% of the 1 mM Mg control. OAA activity showed qualitatively similar metal dependence; those exact percentages were not reported for OAA. At 0.5 mM, phosphate enhanced CHA and OAA activities by 3.0 and 1.7 fold, respectively. These assay observations do not identify the physiological metal. | magnesium(2+)CHEBI:18420 |

- CREATE — Curate the complete eight-member protocatechuate meta-cleavage source definition with individually scoped biological evidence
Follow-up to next253 research and issue534. Added MetaCyc:P184-PWY as a source-defined composite, preserving all eight native reactions, terminal oxaloacetate decarboxylation, exact enol/KCH distinctions, source reversibility, native assemblies, cofactor and regulatory scope, and tested molecular states. Modern LigU/LigJ KCH evidence is not transferred to legacy E,E reactions. Compact source-reading, construct and native-fact dispositions are retained under research/pathway_additions/2026-10-14-next-254/meta-cleavage; original downloads remain external. The transient author-draft catalyst mapping defect is tracked by issue537 and requires independent final replay. This creation event records the curation, not a claim that pending validation or independent review is complete.

- EDIT — Clarify LigK primary assay context and align participant labels with issuing authorities
Follow-up to independent minor source-disposition finding and initial creation history2026-10-14T151353Z-codex-5d93b5.yaml. The Mg cofactor claim remains nonexclusive and supported; added exact CHA metal-relative activity values, qualitative OAA comparison, phosphate enhancement factors, and experimentally qualified recombinant homohexamer context with N-terminal methionine removal. Added PMID12486039 full-source binding and an explicit primary-assay-followup disposition; repeated quotes total22 words for that paper. Seven participant labels now match independent source labels; native IDs, component stoichiometry and all127 graph edges are preserved. Spacing corrections do not modify quotes or source locators. Final independent reassessment remains required.
