nicotine degradation II (pyrrolidine pathway)
MetaCyc:PWY-6993
The complete eleven-step native pyrrolidine pathway converts (S)-nicotine to fumarate and succinate, with methylamine, ammonium and formate coproducts. The experimentally characterized enzymes are scoped to Pseudomonas putida S16. Current NicA2 and Pnao reactions use two oxidized CycN carrier molecules per turnover; their older oxygen-based representations are superseded for this physiological role. N-methylmyosmine hydration is spontaneous. The SpmABC step retains a generic electron acceptor: its physiological acceptor is not assigned. The native NADP-dependent aldehyde and succinate-semialdehyde oxidation steps remain in the complete route, but an exact experimentally supported S16 catalyst is not asserted here for either step. NicA2 assembly reports differ, and functional protein edges do not assert an isolated monomer as the catalytic unit. HspB and maleate isomerase retain verified native dimers. CycN reoxidation, transport and metabolism beyond fumarate, succinate and the released coproducts are outside this record.

- CREATE — Add the complete eleven-reaction nicotine pyrrolidine degradation pathway.
Preserved Pseudomonas putida S16 protein and sequence bridges, native SpmABC identities, CycN-dependent dehydrogenases, full terminal reaction scope and unresolved strain-specific catalyst assignments. Corrected retired ChEBI mapping slots under issue341 without losing replacement provenance. Source and duplicate-search dossiers are in research/pathway_additions/2026-10-11-nicotine-next; shared authorities, generated pages, final gates and independent reviews are recorded separately.
- CREATE — Add the complete eleven-reaction nicotine pyrrolidine degradation pathway.
- Codex
- EDIT — Correct native-source attribution on nicotine catalyst and sequence evidence.
Independent review found an overbroad native catalyst assertion and ChEBI assignments attributed to native pathway ordering. Eleven affected edges now cite the native links plus separate chemistry/replacement sources; nodes and reactions are unchanged. Before/after source receipts and independent reassessment are retained in the cohort dossier.
- EDIT — Correct native-source attribution on nicotine catalyst and sequence evidence.
