fructoselysine and psicoselysine degradation
MetaCyc:PWY0-521
The source-defined three-reaction pathway converts psicoselysine to fructoselysine through FrlC, then uses ATP-dependent FrlD phosphorylation and FrlB deglycation to produce glucose 6-phosphate and lysine. Fructoselysine enters at the kinase step. The two downstream reactions overlap the narrower GO:1901281 fructoselysine catabolic record; these are overlapping boundaries, not disjoint capabilities or exact identifier synonyms. The graph selects catabolic traversal while preserving FrlC reversibility and in vitro FrlB reversibility. Native MG1655 annotations, reviewed K-12 protein identities, experimental constructs and conditional growth evidence retain separate scopes.

Probable fructoselysine/psicoselysine transporter FrlAUniProtKB:P45539experimental construct: Indirect gene-product attribution from the FB21138 frlA Tn5 growth comparison at 20 mM substrates. The Materials passage does not specify this mutant background; the separately described BL21 cultures do not establish it. No complementation or purified FrlA transport assay is reported. Qualification evidence
| enables FrlA participation is inferred from the growth phenotype and reviewed probable-transporter annotation. Tn5 polarity and lack of rescue limit gene-product attribution; transport itself, coupling and stoichiometry were not measured. FrlA is context, not a core reaction member. | MetaCyc:PWY0-521 |

Fructoselysine 6-kinaseUniProtKB:P45543experimental construct: FrlD from BL21(DE3) genomic DNA in pET-3a, expressed in BL21(DE3)pLysS; no affinity tag stated. Qualification evidence
| catalyzes Recombinant FrlD catalyzes ATP-dependent C6 phosphorylation. Product NMR establishes phosphate position while retaining multiple solution forms at pH 4.1. Its start primer matches current P0AC00 residues 1 onward; complete donor sequence identity is unproved. The second-AUG construct was insoluble/inactive. Approximately 450-kDa gel filtration suggests a dodecamer; the assembly trace was not shown. Qualification evidence
| catalyzes Recombinant FrlB supports reversible cleavage to lysine and glucose 6-phosphate, including the reverse condensation experiment. The enzymatic glucose-6-phosphate identification does not establish anomer specificity: the beta product attached to this native reaction remains a separate database assertion. | fructoselysine 6-phosphate + water → beta-D-glucose 6-phosphate + L-lysineMetaCyc:RXN0-963 |

Probable fructoselysine utilization operon transcriptional repressorUniProtKB:P45544experimental construct: E. coli FrlR expressed from pBAD33 in BW25113/JW5698 PfrlABCD-lux reporter comparisons. Exact construction/primer tables were unavailable; full insert identity to current K-12 P45544 is not independently established. The measured target is a shared-promoter reporter, not each endogenous transcript or an MG1655 gene instance. Qualification evidence
| inhibits Transcriptional repression of the shared frlABCD promoter controls pathway expression in the BW25113 reporter system. This process-level context edge does not assert direct inhibition of the pathway enzymes, separate endogenous transcript measurements, regulation of frlR itself, or psicoselysine-specific reporter testing. | MetaCyc:PWY0-521 |

Probable fructoselysine utilization operon transcriptional repressorUniProtKB:P45544experimental construct: Separate in vitro preparation: His6-SUMO-FrlR expressed in BL21(DE3), cleaved with His6-Ulp1 and recovered after metal-affinity cleanup. Exact supplementary construct sequence is unverified. Operator DNA is the assay target; the reference protein ID does not establish a particular MG1655 promoter or full-length native dimer. Qualification evidence
| regulates Operator binding supplies the molecular basis for the separately observed transcriptional repression. SPR measured operator-dependent binding, with reported KD 4.9 nM after active-concentration correction; engineered T7/operator reporters support sequence dependence. This is regulatory context, not a new metabolic event. | MetaCyc:PWY0-521 |

RNA polymerase sigma factor RpoHUniProtKB:P0AGB3experimental construct: pBAD33-rpoH overexpression in BW25113 shared-promoter lux reporter assays, with promoter truncations and temperature conditions specified by Figure 5c and Methods4.5. Exact supplementary insert sequence was not inspected; current K-12 RpoH denotes gene-product identity. Qualification evidence
| activates Positive transcriptional context from RpoH overexpression and promoter-truncation reporters. This is not a purified sigma-factor binding experiment, direct enzyme activation, individual endogenous-transcript measurement, or a host-adaptation experiment. | MetaCyc:PWY0-521 |
|
DNA-binding transcriptional dual regulator CRPUniProtKB:P0ACJ8experimental construct: Indirect CRP attribution in BW25113/JW3778 cyaA and PfrlABCD-lux reporter comparisons. CRP itself was neither deleted nor purified in these experiments; promoter DNA and reporter context were engineered. Exact equivalence of every tested sequence to the reference K-12 molecules is not established. Qualification evidence
| activates Inferred CRP/cAMP-dependent positive transcriptional control from cyaA manipulation, glucose response and candidate-CRP-region truncation. The study supplies no new purified CRP binding or crp-deletion experiment. The source differs between 10 and 20 mM glucose descriptions; no single dose is selected here. | MetaCyc:PWY0-521 |

- CREATE — Curated the complete native fructoselysine and psicoselysine pathway with scoped experimental and regulatory evidence
Added MetaCyc:PWY0-521 with the FrlC entry and shared FrlD/FrlB core. Complete native and Rhea sides retain beta-glucose 6-phosphate under native RXN0-963. Broader Rhea28383 is not an exact substitute. Preserved GO1901281 unchanged. Experimental constructs, strain differences, metal alternatives and promoter-reporter regulation retain explicit scope. Issue514 source-local wording repair was independently checked before subsequent evidence integration. Research, source appraisals, required repository gates and saved scientific review are retained separately with their actual status.
