paraflagellar rod
A large lattice-like axial structure found in some flagellated protists which extends alongside the axoneme. Protein components of the paraflagellar rod are likely implicated, among other, in adenine nucleotide signaling and metabolism, and in calcium signaling. [GO:0097740]
Synonyms: PFR; paraxial rod
Part of: GO:0097729 — 9+2 motile cilium (no local record)
Components
| Component | Type | Part of / acts on | Grounding | Genes | Stoichiometry | Essential | Role |
|---|---|---|---|---|---|---|---|
| paraflagellar rod protein PFR1 | Protein | part of | Reviewed label only (reviewed name; no accession asserted) | PFR1, PFRC, PFRD | ESSENTIAL | Major lattice protein that co-assembles with PFR2 in the full paraflagellar rod of the studied trypanosome and Leishmania models; Leishmania PFR1 can associate with axoneme attachment fibers without PFR2.
| |
| paraflagellar rod protein PFR2 | Protein | part of | Reviewed label only (reviewed name; no accession asserted) | PFR2, PFRA, PFRB | CONDITIONAL | Major lattice protein required for full native PFR architecture in the studied trypanosome and Leishmania models, not for every reduced PFR. Gadelha et al. found no evidence for PFR2 in the reduced Crithidia deanei rod using several assays, while noting their detection limitations.

Taxonomic distribution
NCBITaxon:2759Eukaryota — RARE: The paraflagellar rod is restricted to a small set of flagellated protist lineages rather than a conserved feature of eukaryotic cilia. [PMID:7961827]NCBITaxon:33682Euglenozoa — VARIABLE: Within Euglenozoa, PFRs are prominent in kinetoplastid flagella, but they can be greatly reduced and cryptic in some endosymbiont-bearing kinetoplastids. Functions
- flagellar waveform control — The PFR provides an extra-axonemal scaffold needed for normal kinetoplastid flagellar beating and cell motility.
DOI:10.1038/35300Bastin et al. 1998 disrupted the PFR in T. brucei and linked PFR loss to a trypanosome motility defect.PMID:9497035Santrich et al. 1997 showed that a Leishmania mexicana PFR2 knockout lacks most PFR structure and has altered flagellar beat waveform and forward swimming.
- extra-axonemal PFR assembly — New PFR lattice proteins are delivered into the flagellum and incorporated into the elongating paraflagellar rod, a process that requires cytoplasmic PFR assembly factors.
DOI:10.1128/MCB.19.12.8191Bastin et al. 1999 followed tagged PFRA during PFR growth and observed a major distal addition site plus minor incorporation along the partially assembled PFR.DOI:10.1242/jcs.242271Alves et al. 2020 identified cytoplasmic PFR-AF1 and PFR-AF2 as factors required for PFR formation.
- flagellar signaling platform — The PFR-associated protein cohort includes calcium/calmodulin and adenine-nucleotide signaling or metabolic proteins that can influence flagellar beating.
DOI:10.1074/jbc.M808859200Portman et al. 2009 used RNAi-mutant comparative proteomics to expand the PFR cohort and infer calcium/calmodulin regulatory links among some components.DOI:10.1016/j.ijpara.2009.10.005Portman and Gull 2010 review links between the PFR proteome, adenine-nucleotide metabolism, calcium signaling and motility regulation.
Mechanism graphs
PFR1 and PFR2 build the paraflagellar rod lattice (ASSEMBLY)
PFR1 and PFR2 co-assemble into the full PFR lattice in the studied Trypanosoma and Leishmania models. In T. brucei, cytoplasmic PFR-AF1 and PFR-AF2 act upstream of flagellar assembly and support major PFR protein accumulation. This model does not require the same composition in reduced rods such as that of Crithidia deanei.

Discussions
- CURATION_TODO (OPEN): Should all proteomic PFR candidates be curated as paraflagellar-rod components now? The PFR proteome contains many low-abundance or regulatory proteins in addition to the two major lattice proteins; Portman et al. 2009 identified 20 novel PFR components by RNAi-ablation proteomics and epitope tagging. This first GO:0097740 record therefore enumerates only PFR1/PFR2, while additional candidates need family-by-family curation to avoid folding transient or nearby flagellar proteins into the structural lattice boundary.
- CURATION_TODO (RESOLVED): Are PFR-AF1 and PFR-AF2 paraflagellar-rod components? PFR-AF1 and PFR-AF2 are cytoplasmic proteins required for PFR formation, but the component boundary in docs/CURATION.md excludes factors that act on a subunit before assembly and then depart.
- INTERPRETATION (RESOLVED): Reconcile historical PFRA/PFRC and UniProt names with cross-species PFR1/PFR2 orthology. Gadelha et al. 2004 Table 1 and Fig. 1 consolidate T. brucei PFRC/PFRD (Q26789, 73 kDa) as PFR1 and PFRA/PFRB (P22225, 69 kDa) as PFR2. Historical UniProt mnemonics use the opposite ordinal numbering and must not substitute for this cross-species family assignment.
