prostheca
A tubular projection of the bacterial cell envelope and cytoplasm that extends away from the main cell body in prosthecate bacteria. In stalked Caulobacter cells, the prostheca contains the inner membrane, peptidoglycan and outer membrane layers of the Gram-negative envelope and supports stalk-specific protein localization. Functions
- polar envelope extension — Cell-cycle-coupled stalk biogenesis extends the Caulobacter envelope at the old pole after the swarmer-to-stalked transition.
DOI:10.1111/j.1365-2958.2005.04970.xBiondi et al. 2006 identify a phosphorelay system that controls Caulobacter stalk biogenesis during cell-cycle progression.
- stalk nutrient capture and hydrolysis — Caulobacter stalks can take up and hydrolyze the organic phosphate substrate fluorescein diphosphate. Modeling predicts that a long, thin stalk can improve diffusive nutrient capture, but delivery of captured nutrients to the cell-body cytoplasm and the resulting nutritional benefit remain unresolved.
DOI:10.1073/pnas.0602047103Wagner et al. 2006 directly observed uptake and hydrolysis of fluorescein diphosphate by attached and isolated stalks. Their diffusive-capture model predicts a length/geometry advantage, not direct proof of whole-cell nutritional benefit.DOI:10.1046/j.1365-2958.2002.03071.xIreland et al. 2002 identified transport-related proteins in isolated stalks, consistent with uptake competence but not proof of nutrient delivery to the cell body.DOI:10.1016/j.cell.2012.10.046Schlimpert et al. 2012 show that mature crossbands block PstS protein exchange, challenging the proposed PstS-mediated phosphate shuttle. Possible small-molecule routes were not established.DOI:10.1371/journal.pone.0184063Curtis 2017 discusses the unresolved Caulobacter nutritional role and shows PstA-GFP in some Brevundimonas stalks; that localization does not demonstrate a complete transporter or justify a shared mechanism across species.
- stalk compartmentalization — Protein diffusion barriers divide the Caulobacter stalk from the cell body and separate stalk segments from each other.
DOI:10.1016/j.cell.2012.10.046Schlimpert et al. 2012 show that crossband diffusion barriers compartmentalize the Caulobacter stalk.
Mechanism graphs
MreB-dependent wall synthesis extends the Caulobacter stalk (ASSEMBLY)
Cell-cycle regulators control Caulobacter stalk biogenesis, MreB-dependent stalk-wall machinery builds stalk-specific peptidoglycan, and continued envelope extension yields a polar prostheca whose StpX and crossband proteins help maintain local stalk organization.

Discussions
- CURATION_TODO (RESOLVED): Keep Caulobacter-specific StpX, crossband proteins and stalk wall-synthesis proteins out of the prostheca component table. This record denotes the envelope extension, whose cross-lineage constituents are the wall and membrane materials, not every protein needed for Caulobacter stalk elongation or compartmentalization. The first mechanism graph captures PbpC/MreB-dependent stalk wall synthesis, StpX localization and StpABCD crossbands as Caulobacter actors until exact component boundaries can be asserted.
- CURATION_TODO (OPEN): Resolve exact source-neutral groundings for stalk-specific peptidoglycan and stalk envelope lipids. The prostheca is built by extension of chemically diverse envelope layers, so the initial record uses reviewed labels instead of over-specific generic chemical CURIEs.
- INTERPRETATION (OPEN): Distinguish demonstrated stalk substrate capture from an unresolved whole-cell nutritional role. Wagner 2006 demonstrated FDP uptake/hydrolysis and modeled diffusive capture, but later crossband experiments block the proposed PstS shuttle toward the cell body. Small-molecule delivery and surface-elevation benefits remain hypotheses; Brevundimonas PstA localization does not resolve the Caulobacter transport route. The graph therefore records the measured substrate-specific activity rather than a settled whole-cell nutrient mechanism.
