stressosome

cellstructuremech:stressosome · Other · Multiprotein complex · Proposed

A bacterial multiprotein regulatory assembly with an oligomeric core formed by RsbR-family and RsbS STAS domains and an associated RsbT kinase. Characterized stressosomes organize environmental-stress signaling; their subunit composition and requirement for downstream responses depend on the organism and experimental context. [DOI:10.1038/s41467-019-10782-0]

Reconstituted Listeria stressosome particles: SDS-PAGE (a), negative-stain EM comparison of RsbRST and RsbRS preparations (b), cryo-EM micrograph (c), reconstructed density (d), and local density/model comparisons (e). These are purified particles, not an in-cell localization image; no atomic RsbT model is resolved.

The RsbR paralog repertoire and sensory-domain type vary between organisms.
  • DOI:10.1038/s41467-019-10782-0 Listeria cryo-EM resolves RsbR STAS domains in the core; the N-terminal turrets are less well resolved.
  • DOI:10.1107/S205225251900945X Bacillus RsbRA and RsbS form reconstituted cores; the paper distinguishes the LOV-domain paralog YtvA from globin-like RsbR proteins.

RsbTProteinpart ofReviewed label only (reviewed name; no accession asserted)rsbTUNKNOWNAssociated signaling kinase in RsbR-RsbS-RsbT particles. Its necessity for downstream signaling is distinct from its necessity for formation of an RsbR-RsbS core.
Stressosome-associated protein Prli42Proteinacts onInterPro:IPR049722prli42DISPENSABLEListeria membrane-associated miniprotein implicated in RsbR-dependent stressosome tethering, not a generic core subunit. Dispensability refers to reconstituted RsbR-RsbS-RsbT assembly without Prli42, not normal localization or stress signaling in vivo.
  • DOI:10.1038/nmicrobiol.2017.5 EGD-e co-immunoprecipitation identifies RsbR and RsbS among Prli42 partners; deletion and partial complementation support Prli42-dependent membrane association of a subset of RsbR. Fractionation measures RsbR, not counts of intact stressosomes. Purified RsbR-RsbS-RsbT particles assemble without Prli42.
  • InterPro:IPR049722 The issuing authority identifies an exact Prli42 protein family; the family label does not establish occurrence of assembled stressosomes in every organism with a homolog.
